Mechanism of Action

Itraconazole is a synthetic triazole antifungal that exerts its fungistatic and fungicidal activity by selectively inhibiting the cytochrome P450-dependent enzyme lanosterol 14-alpha-demethylase (CYP51) in fungal cell membranes. This enzyme catalyses the conversion of lanosterol to ergosterol — the essential structural sterol of the fungal cell membrane. Without ergosterol, the fungal membrane loses its structural integrity, becoming abnormally permeable, functionally impaired, and unable to maintain the osmotic balance and transport functions essential for fungal viability.

Indications

Dermatological Fungal Infections (Primary Indication): Dermatophytosis (tinea corporis, tinea cruris, tinea pedis, tinea manuum), pityriasis versicolor, and tinea capitis. Oropharyngeal and Oesophageal Candidiasis: In patients intolerant of fluconazole or with fluconazole-resistant Candida.
